A question about Plug-in

My question is if I have 2 different card linked in a system, and one card have Plug-in Package A and another card has Plug-in package B, would both plug-in packages work within the system setup? Or plug-in only goes in function with the main card you use in your setup?

hehe, I know this question may sound a bit stupid. It does make more sense to work that well as well, but I just wanted to confirm it and not make too many assumptions
